Hydroxypropyl Cyclodextrin Improves Amiodarone-induced Aberrant Lipid Homeostasis of Alveolar Cells

چکیده

Alveolar epithelial type II (AT2) cells secrete pulmonary surfactant via lamellar bodies (LBs). Abnormalities in LBs are associated with disorders, including fibrosis. However, high-content screening (HCS) for LB abnormalities is limited by the lack of understanding AT2 cell functions. In present study, we have developed harboring LB-like organelles that proteins. These were more similar to than parental A549 cells. recapitulated amiodarone (AMD)-induced enlargement, patients exposed AMD. To reverse AMD-induced abnormalities, performed HCS approved drugs and identified 2-hydroxypropyl-?-cyclodextrin (HP?CD), a cyclic oligosaccharide, as potential therapeutic agent. A transcriptome analysis revealed HP?CD modulates lipid homeostasis. addition, inhibited human induced pluripotent stem cell–derived Our results demonstrate useful suggest candidate agent interstitial pneumonia.

The Effects of Amiodarone of Rabbit Lung Alveolar Cells

Purpose: The effects of Amiodarone on lung tissue and morphologic changes of alveolar cells. Materials and Methods: in this study, we used 21 mature male rabbits and they were divided into control and two experimental groups. Daily 80 mg/kg amiodarone was injected intra peritoneally to the experimental animals for one or two weeks. The control group received normal saline the same dose as amio...

Rituximab therapy in pulmonary alveolar proteinosis improves alveolar macrophage lipid homeostasis

RATIONALE Pulmonary Alveolar Proteinosis (PAP) patients exhibit an acquired deficiency of biologically active granulocyte-macrophage colony stimulating factor (GM-CSF) attributable to GM-CSF specific autoantibodies. PAP alveolar macrophages are foamy, lipid-filled cells with impaired surfactant clearance and markedly reduced expression of the transcription factor peroxisome proliferator-activat...

Safety of Parenteral Hydroxypropyl /?-Cyclodextrin

0 Post-treatment data were collected on a patient who received intravenous hydroxypropyl P-cyclodextrin in a dose of 1.5 g/kg in 1985. Although no untoward effects were observed in this patient, rarely occurring agitation and pulmonary edema have been noted after injections into rabbits and dogs, respectively. These complications are analyzed here on the basis of symptoms and on the effects of ...

Altered surfactant homeostasis and alveolar epithelial cell stress in amiodarone-induced lung fibrosis.

Amiodarone (AD) is a highly efficient antiarrhythmic drug with potentially serious side effects. Severe pulmonary toxicity is reported in patients receiving AD even at low doses and may cause interstitial pneumonia as well as lung fibrosis. Apoptosis of alveolar epithelial type II cells (AECII) has been suggested to play an important role in this disease. In the current study, we aimed to estab...

Development and Characterization of Paracetamol Complexes with Hydroxypropyl-?-Cyclodextrin

Paracetamol is a sparingly soluble bitter tasting drug. It is widely used as an analgesic and antipyretic. Complexation of drug with different cyclodextrins (?, ? and HP-?-CD) was attempted to improve solubility of Paracetamol. During the drug excipient interaction studies, ?, ? cyclodextrins elicited analytical interference and showed considerable absorbance at ?max (243.5 nm) of Paracetamol w...
